CALLUS INDUCTION AND PLANTLET REGENERATION IN CHRYSANTHEMUM

The experiment was carried out to study the in vitro indirect regeneration in chrysanthemum during the period of February 2016 to June 2017 at Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-e-Bangla Nagar, Dhaka-1207. MS medium supplemented with different concentrations and combinations of BA, 2,4-D and IAA were used for callus induction and plantlet regeneration. Nodal segments were used as explants. The minimum (14.20 days) for highest frequency (76%) of callus were recorded in BA 2.0 mg/l + 2,4-D 1.0 mg/l treatment whereas; no callus was observed in control treatment. The treatment BA 2.0 mg/l + 2,4-D  1.0 mg/l gave the highest no. of shoot (3.20) within minimum days (9.20).Highest percentage of root within minimum days was recorded by 0.5 mg/l of IAA whereas; higher doses of IAA such 1.0 mg/l and 1.5 mg/l showed the best response in case of  length (3.16 cm) and number (5.40) of root respectively. The control treatment gave the poor results for most of the parameters of root.
